[摘 要]采用超声波辅助草取法提取,高效液相色请法测定了内器(Cinhe)中的儿茶素含量。在单因素试验基础上,以料液比、提取温度、超声时间、提取次数为研究对象,进行了4因素3水平正交试验。结果表明内欢蓉中儿茶素最佳提取工艺为。甲醇料液比2g:25ml,提取温度60℃,超声提取30min,提取1次。儿茶素在0.602-15.05 pghL.范围内呈良好线性关系,回归方程为y-19300x+421095.1-0.9996.加样回收率为98.25%。在条件下,肉成蓉中儿茶素含量为25.77mg/g。

Study on Extraction Conditions and Content Determination of Catechin from
Cistanche

Abstractt Catechin in Citanche was alrasonically extracted and detectod by HPLC.Acconding o single-factor experiments,three level of four factors incliting material -lkguid ratio,uhravonic emperature,alrasonic tme and ulrasonic mumber were sleced for the orthogonal design.The resuls showed that the optimum extraction condtions as follows:makrialiqud ratio was2g:25ml nih 30 mn ulrasonicansiod extaction under 60 C,with anlyone ultrasound was performed.Uinder the aptimal conditions,the cahbration cure of Chiectin was linca(-0.9996)in the range of 0.502-15.05 ughl,The equation was y-19300x+421095,-0.9998,the recovery was 98.26 %and the conent of Catectin from Catnche was 26.77 mg/g This process can effectively extrat catechin from cistanche,and provide a bavis for the improvement of pharmacopocia sandard in the fture.

肉苁蓉(Catanche deserticola YC.Ma)是我国中医用途最广的珍贵中草药之一,习称大芸、苁蓉、为列当科(Oobanchancae)肉苁蓉属(Cistanche)多年生寄生草本植物,专性寄生在藜科(Chenopodiaceae)校梭(Haloncylon ammodendon(C.A.Mey)Bunge) 和白梭梭(H.Persicum Bungeex Boss)的根部。主产于我国内蒙、新疆、甘肃、宁夏、青海等省(区)以及伊朗、蒙古、印度等国。其味甘咸微辛酸、微温。在《神农本草经》中将其列为上品,中国药真规定正品为干燥带鳞叶的肉质。肉苁蓉具有强肝肾。益精血,滑肠通便之功效,动物实验显示有降压作用,其中所含儿茶素具有提高免疫功能、清除自由基、抗氧化作用、防突变。抗癌等活性,与肉苁蓉主要药理作用相吻合。本课题经 HPLC预试验发现儿茶素含量较高,但中国药典对其含量的测定方法却没有具体标准,面对肉苁蓉中儿茶素的含量测定目前尚未见有文献报道。因此,本文将对肉苁蓉中儿茶素的提取及含量测定进行条件优化及探索,希望找到一条切实可行的测定方法,为今后肉苁蓉的进一步研究开发提供依据。
